Classical Swine Fever Virus Antibody Test Kit (CSFV Ab)
概述
豬瘟病毒(CSFV)、牛病毒性腹瀉性病毒(BVDV)以及邊界?。˙DV)都屬于黃病毒屬的成員。由于CSFV的高致病力和高致死率,它已給養(yǎng)殖業(yè)造成了巨大的損失。豬感染高致病力毒株后在潛伏期就可以造成傳染,經(jīng)歷急性豬瘟或亞急性豬瘟臨床癥狀的豬在出現(xiàn)癥狀前大量排毒,但耐過后的豬只體內(nèi)有明顯的CSFV抗體,且不再排毒。感染溫和性毒株的豬呈慢性感染,在病死前會長期或間歇性地排毒。懷孕母豬可以通過胎盤感染胎兒導致流產(chǎn)、木乃伊化或產(chǎn)出弱豬、畸形胎等。先天性感染低毒力病毒的結(jié)果是產(chǎn)出持續(xù)性感染的仔豬,仔豬出現(xiàn)免疫耐受,不表現(xiàn)任何癥狀向外排毒。豬也有可能感染BVDV或BDV。盡管這些感染通常呈溫和過程并且是自限性的,但有效的將它們同豬瘟病毒區(qū)分是很重要的。
原理
豬瘟病毒抗體ELISA檢測試劑盒是用來檢測豬血清或血漿中豬瘟病毒抗體的檢測試劑盒。該試劑盒是用豬瘟病毒抗原包被的微量反應(yīng)板,利用阻斷ELISA原理來檢測豬血清或血漿中豬瘟病毒抗體。如果被檢樣品中存在豬瘟病毒抗體,它們就會阻斷辣根過氧化物酶標記(HRPO)的抗豬瘟病毒的單克隆抗體。單克隆抗體與豬瘟病毒抗原的結(jié)合可以通過辣根過氧化物酶與底物的顯色程度進行判定,即用酶標儀在單波長450nm或雙波長450nm與620nm測定該反應(yīng)體系的吸光度。當被檢樣品中含有豬瘟病毒抗體(陽性結(jié)果)時,顯色就會變淺,當被檢樣品中不含有抗豬瘟病毒抗體(陰性結(jié)果)時,顯色就會變深。樣本的阻斷率可以通過450nm波長樣本吸光度與陰性對照吸光度的比值來確定。
The PrioCHECK? CSFV Antibody 2.0 ELISA is a blocking ELISA that specifically detects antibodies against the A domain of the E2 protein of CSFV. This domain is more specific for CSFV than the B, C, and D domain of the E2 protein. An important advantage of the test is that it does not cross-react with related viruses such as Bovine Viral Diarrhoea virus (BVDV) and Border Disease virus (BDV). The test detects antibodies against high-, moderate-, and low-virulent CSFV strains early after infection. Detection of antibodies elicited by low-virulent CSFV strains is of paramount importance to trace sub clinical CSFV infections.
Test procedure
The PrioCHECK? CSFV Antibody 2.0 ELISA is a blocking ELISA. In this test, the plates are coated with the E2 CSFV envelope protein. The key reagent is a monoclonal antibody (mAb) directed against an epitope located on the A domain of the envelope protein E2 of CSFV, making the test more specific for CSFV. The mAb is labeled with an enzyme that generates a color signal. The signal is measured and when no color is formed, the sample containing anti-CSFV antibodies that competed for the A domain of the E2 protein is positive for CSFV.
