表面配體為烷基的ZnSe/ZnS納米熒光探針,硒化鋅-硫化鋅納米熒光量子點(diǎn),具有以下優(yōu)勢:
(1)具有寬的激發(fā)波長范圍和窄的發(fā)射波長范圍,即可以使用小于其發(fā)射波長10 nm的任意波長的激發(fā)光進(jìn)行激發(fā),這樣就可以使用同一種激發(fā)光同時(shí)激發(fā)多種量子點(diǎn),發(fā)射出不同波長的熒光,因而可用于多種標(biāo)記物的同時(shí)檢測,極大地促進(jìn)了熒光標(biāo)記在生物醫(yī)學(xué)中的應(yīng)用。而傳統(tǒng)的有機(jī)熒光染料的激發(fā)光波長范圍較窄,需要多種波長的激發(fā)光來激發(fā)多種熒光染料,這樣給實(shí)際的研究工作帶來了很多的不便。
(2)量子點(diǎn)的發(fā)射峰窄而對稱,且連續(xù)分布,重疊小,這樣,在一個(gè)可檢測到的光譜范圍內(nèi)可同時(shí)使用多個(gè)探針,而發(fā)射光譜不出現(xiàn)交疊,使生物分子的多組分分析檢測變得容易。而傳統(tǒng)的有機(jī)熒光染料的發(fā)射峰不僅寬,而且不對稱,拖尾嚴(yán)重,互相重疊嚴(yán)重,容易互相干擾,給分析檢測帶來難以解決的難題。
(3)量子點(diǎn)的發(fā)射波長可通過控制它的大小和組成來調(diào)諧,可以任意合成所需波長的量子點(diǎn),大小均勻的量子點(diǎn)譜峰為對稱高斯分布,而傳統(tǒng)的有機(jī)熒光染料的峰形則為對數(shù)正態(tài)分布。
(4)量子點(diǎn)的熒光強(qiáng)度比常用的有機(jī)熒光染料羅丹明6G染料高20倍,它的穩(wěn)定性更是羅丹明6G染料的100倍以上,而且量子點(diǎn)抗光漂白能力強(qiáng)。所謂光漂白是指由光激發(fā)引起發(fā)光物質(zhì)分解而使熒光強(qiáng)度降低的現(xiàn)象。有機(jī)熒光染料的光漂白速率很快,而量子點(diǎn)的光漂白作用則遠(yuǎn)遠(yuǎn)小的多,因此可以對所標(biāo)記的物體進(jìn)行長時(shí)間的觀察,并可以毫無困難的進(jìn)行相關(guān)界面的修飾和連接,而不像傳統(tǒng)的有機(jī)熒光染料那樣容易發(fā)生熒光淬滅。這也為研究細(xì)胞中生物分子之間長期相互作用提供了有力的工具。
(5)生物相容性好,尤其是經(jīng)過各種化學(xué)修飾之后,可以進(jìn)行特異性連接,細(xì)胞毒性低,對生物體危害小,可進(jìn)行生物活體標(biāo)記和檢測,而傳統(tǒng)的有機(jī)熒光染料一般毒性較大,生物相容性差。
(6)熒光壽命長,典型的有機(jī)熒光染料的熒光壽命僅為幾納秒(ns),這與很多生物樣本的自發(fā)熒光衰減的時(shí)間相當(dāng)。而量子點(diǎn)的熒光壽命可持續(xù)長達(dá)數(shù)十納秒(20 ns~50 ns),這使得當(dāng)光激發(fā)數(shù)納秒以后,大多數(shù)的自發(fā)熒光背景已經(jīng)衰減,而量子點(diǎn)熒光仍然存在,此時(shí)即可獲得無背景干擾的熒光信號。
這些*的光學(xué)特性,使量子點(diǎn)成為了一種理想的熒光探針。因此,使用量子點(diǎn)代替有機(jī)熒光染料,將在細(xì)胞定位、信號傳導(dǎo)、細(xì)胞內(nèi)分子的運(yùn)動(dòng)和遷移等研究中發(fā)揮重要的作用。鑒于以上這些優(yōu)點(diǎn),量子點(diǎn)的制備以及在各領(lǐng)域的應(yīng)用研究都取得了突破性進(jìn)展。
