簡介:
琥珀酸是半胱氨酸殘基的一種穩(wěn)定的翻譯后修飾,其會改變蛋白質功能或轉變以響應細胞內氧化還原環(huán)境的變化。當Krebs循環(huán)中間體富馬酸鹽與半胱氨酸反應產生S-(2-琥珀酸)半胱氨酸(2SC)時,就會發(fā)生丁二酸。大量的蛋白質會發(fā)生琥珀酸化,包括酶、脂肪因子、細胞骨架蛋白和ER伴侶,琥珀酸化已被證明在調節(jié)生物學中發(fā)揮作用。在糖尿病中可以看到脂肪細胞蛋白琥珀化的增加,這是由營養(yǎng)過剩引起的線粒體應激引起的。因此,2SC被作為慢性疾?。ㄈ绶逝?、糖尿病和癌癥)中線粒體應激或功能障礙的生物標志物。
