納米金Gold Nanoparticles的制備方法主要采用還原劑,如檸檬酸鈉,硼氫化鈉等還原氯金酸HAuCl4·4H2O。氯金酸HAuCl4·4H2O在還原劑的作用下,可以聚一定大小的金納米顆粒,形成帶負(fù)電的疏水膠溶液。由于靜電作用而成為穩(wěn)定的膠體狀態(tài),故稱膠體金Colloidal gold。
納米金隨粒徑的變化而顏色發(fā)生變化:不同尺寸形貌的納米金對可見光有選擇性吸收,吸收了某個波長的光后,將沒有吸收的光反射出去,我們看到的就是反射的光,其實不是他們的顏色,不過是他們將這種顏色的光反射給我們看到了,這種光在我們的視覺中是某種顏色而已。不同尺寸形狀的顆粒,它們的阻尼現(xiàn)象不同,一般來說小顆粒的阻尼現(xiàn)象,從而小顆粒振動頻率要小于大顆粒,它們也就吸收長波的光,反射短波的光。隨著顆粒增加,它們吸收的光波波長變短,也就是所謂的紅移現(xiàn)象了。

2納米金應(yīng)用于蛋白質(zhì)纖維染色的研究
納米金溶膠應(yīng)用于對羊絨、羊毛纖維及羊毛織物進行染色,探討了納米金與蛋白質(zhì)的作用原理。(早孕試紙:HCG膠體金法檢測試劑盒采用雙抗體夾心一步法技術(shù),以膠體金為指示標(biāo)記,檢測尿液中的HCG濃度)
3 納米金應(yīng)用于的研究進展
金納米粒子具有的生物親和性及的表面性質(zhì),經(jīng)表面修飾的金納米粒子通過共價或非共價的方式與靶向分子或者相連,獲得多功能納米粒子,應(yīng)用于的靶向與。金納米粒子是一種的靶向制劑,可以同時實現(xiàn)被動靶向和主動靶向:有納米粒徑大小引起的通透性和滯留效應(yīng)(enhanced permeability and retention effect , EPR )可以實現(xiàn)被動靶向,使納米金載體在組織選擇性分布,以增加并減少副作用。此外,金納米粒子進行表面修飾后,可利用靶向分子實現(xiàn)的主動靶向,將或射線特異帶到部位,靶向的目的。納米金團簇表面的邊緣位點和頂部位點4f軌道的結(jié)合能相對較低,這些位點形成的Au-S鍵的鍵能也相對較低,容易發(fā)生離解,這對Au-MMPCs的穩(wěn)定性和配體交換有影響.Au-MPCs具有粒徑小和生物相容性好的,用巰基或二硫化合物對Au-MPCs表面進行修飾,形成具有生物活性的Au-MMPCs,這些生物活性Au-MMPCs同待轉(zhuǎn)運物和膜相互作用,通過靜電吸附和內(nèi)吞作用可將轉(zhuǎn)運物傳送和核內(nèi),靶向的傳輸和釋放需要依靠Au-MPCs的作用.
4 納米金生物探針及其應(yīng)用
納米金顆粒的表面不表現(xiàn)為靜電作用,還有疏水作用。為了讓蛋白質(zhì)與納米金顆粒結(jié)合,減小靜電作用,而是疏水作用起作用。通過表面改性,讓納米金帶正電,從而與帶負(fù)電的蛋白質(zhì)相結(jié)合,制成用于電子顯微鏡的金探針。膠體金與抗人球蛋白(anti human-IgC)結(jié)合制備的金探針可以定性檢測艾滋(HIV)抗體。在納米金微粒上修飾DNA探針,由于共價結(jié)合力,在DNA探針的一段連接上巰基后可與納米金結(jié)合,通過識別顏色的變化,就能檢測出相應(yīng)的靶DNA。
5納米金在生物標(biāo)記分析中的應(yīng)用進展
金納米粒子除具有一般納米粒子的表面效應(yīng),量子尺寸效應(yīng)外,同時具有的生物相容性。由于生物大分子和功能化同處于納米維度當(dāng)在相應(yīng)的生物體系中加入功能化的納米金顆粒使其具有定向吸附效應(yīng)以及富集效應(yīng)等。兩者通過化學(xué)或物理吸附可制得既穩(wěn)定又具有一定計量關(guān)系的生物/納米復(fù)合材料。該復(fù)合材料又可以用來構(gòu)建具有功能的分子結(jié)構(gòu)體系,此外也可以作為檢測探針用于研究復(fù)雜的生物化學(xué)體系。
6 納米金在生物傳感器中的應(yīng)用
膠體金增DNA生物傳感器的檢測靈敏度,納米級的金顆粒標(biāo)記在DNA分子上,可增加該分析分子的質(zhì)量,從而增質(zhì)量性。膠體金能提供一似于能保持蛋白質(zhì)穩(wěn)定性的,實習(xí)附在其表面的蛋白質(zhì)保持他們的生物活性,能增乳酸生物傳感器的檢測靈敏度。用金納米粒子修飾鉑電而制成的葡萄糖氧化(GOD)生物傳感器,具有靈敏度,速度快等。
